Ralph (Raphe) Scrope, esq. of Hambleden, Bucks was the eldest son of John Scrope, second son of Sir Henry Scrope, 6th Baron Scrope of Bolton. He was born in 1518 in Hambleden, Buckinghamshire, England. He married in about 1564 Lady Elizabeth, daughter of William, Lord Windsor of Stanwell, Middlesex, England. They had one daughter and three sons:
